Definition

What is a stable isotope-labeled compound?

It is a molecule in which selected atoms are enriched with non-radioactive isotopes such as deuterium (²H), carbon-13 (¹³C), nitrogen-15 (¹⁵N) or oxygen-18 (¹⁸O). The labelled molecule can be differentiated by mass or spectroscopy while retaining close chemical similarity to its unlabelled counterpart.

Isotope choice

The label must fit the method

Deuterium

Often provides an accessible mass shift. Position and hydrogen-deuterium exchange must be considered.

Carbon-13

A non-exchangeable label commonly used when close chromatographic behaviour and label stability are important.

Nitrogen-15

Useful in nitrogen-containing molecules, metabolic tracing and multi-isotope internal-standard designs.

Oxygen-18

Used in selected reaction, biochemical and tracer studies; incorporation and exchange conditions require review.

Multiple labels

Combined isotopes can increase mass separation or place a durable label at a structurally suitable site.

Unlabelled counterpart

The analyte structure, ion fragments, matrix, method and possible interference all influence the best standard.
